5-Methyltetrahydrofolic Acid, 5-Formyltetrahydrofolic Acid (Folinic Acid), and Folie Acid Requirements of Normal and Rous Sarcoma Virus-infected Chicken Fibroblasts1

چکیده
Normal and Rous sarcoma virus-infected chicken fibroblasts proliferate maximally in a culture medium contain ing a physiological (10 ng/ml) concentration of 5-methyltetrahydrofolic acid or folinic acid (5-formyltetrahydrofolic acid), while their maximal proliferation requires a hyperphysiological (1000 ng/ml) concentration of folie acid. The normal and Rous-infected fibroblasts do not differ in their requirements for 5-methyltetrahydrofolate, folinic acid, or folie acid.

Single (13)C6-labelled doses of pteroylmonoglutamic acid (PteGlu; 634 nmol) or 5-formyltetrahydrofolic acid (431-569 nmol) were given to fasted adult volunteers, and the rise in total and (13)C-labelled plasma 5-methyltetrahydrofolic acid metabolite monitored over 8 h by HPLC and liquid chromatography-MS. The dose-adjusted area under the curve (AUC) for total (labelled plus unlabelled) plasma 5...
متن کاملCell cycle-dependent activation of rous sarcoma virus-infected stationary chicken cells: avian leukosis virus group-specific antigens and ribonucleic acid.
Stationary chicken embryo fibroblasts exposed to Rous sarcoma virus (RSV) remained stably infected for at least 5 days, but they did not release infectious virus or become transformed until after cell division. These infected stationary cells did not contain avian leukosis virus group-specific antigens or ribonucleic acid (RNA) hybridizable to deoxyribonucleic acid (DNA) made by the RSV endogen...

Comparisons were made of the rate of incorporation of C'4-labeled L-arnino acids into protein of chicken embryo chorioallantoic membrane (CAM) infected with viruses that induce cell proliferation. A tumor virus (Rous sarcoma virus) and a nontumor virus (fowl pox virus) were used in these studies. In ovo studies of glycine-C14 incor poration into the protein of Rous sarcoma virus-induced tumors ...
متن کاملRoles of calcium, serum, plasma, and folic acid in the control of proliferation of normal and Rous sarcoma virus-infected chicken fibroblasts.
In a culture medium of pH 7.4 and a folic acid concentration of 100 mug/liter that contains 5% heat-inactivated chicken plasma rather than serum, the rate of proliferation of normal chicken fibroblasts is determined by the concentration of calcium. Proliferation, rapid when the calcium concentration is physiological, decreases when the calcium concentration is reduced. At a very low calcium con...
